A resistance training review keeps MGF in the IGF-1 exercise story
A 2026 review of resistance training and nutrition names MGF as one of the local IGF-1 isoforms involved in exercise adaptation. Two other newly indexed studies cover liver and anti-inflammatory pathways, not MGF itself.

A new batch of research tagged with MGF is mostly not about MGF. One paper, a 2026 review of resistance training and nutrition, names mechano growth factor as one of the local IGF-1 isoforms involved in the body's response to exercise 1. The other two papers cover a mangiferin salt for alcoholic liver disease and a ginger-processed herb for inflammation, and neither studies MGF 2. The direct evidence for the MGF research summary still comes from older cell and animal work.
What the resistance training review says
The review pulled together 26 research articles on how resistance training variables and nutrition influence the IGF-1 axis in sedentary people 1. It places MGF among the local IGF-1 isoforms that are part of that axis, but it does not report new MGF measurements 1. The main finding is that low-load training, around 20 to 30 percent of a person's one-repetition maximum, can activate the mTOR pathway, a cellular growth pathway that controls protein synthesis, as effectively as traditional high-load training when blood flow restriction or slow-motion techniques are used 1. For beginners, low-load protocols with blood flow restriction or slow-motion work tap the same mTOR pathway as heavy lifting 1. The review also notes that low-load methods may reduce the roughly 20 percent drop in arterial compliance often linked to heavy lifting 1. Longer rest intervals of about three minutes and evening workouts are described as supportive for adaptation 1. The review also reports that during exercise, up to 88 percent of cardiac output can be redirected to skeletal muscle 1. A nutrigenetic detail is that people carrying the T version of a common gene variant called rs6214 showed greater visceral fat loss when resistance training was paired with daily dairy consumption 1.
What the other studies found
The first unrelated study tested mangiferin monosodium salt, a compound abbreviated MG-Na, in rats with alcohol-induced liver disease 2. It found that MG-Na improved liver function and intestinal barrier integrity and shifted the diversity and richness of gut microbiota 2. The study also reported changes in a copper-dependent form of cell death called cuproptosis, plus lower levels of inflammatory signals such as IFN-beta, TNF-alpha, IL-1 beta, and IL-6 2. The MG-Na treatment downregulated proteins involved in cuproptosis, including FDX1 and DLAT, and reduced cGAS-STING and TLR4/NF-kB/NLRP3 pathway activity 2. It also reduced fecal metabolites such as (R)-lipoic acid and pyruvic acid in the alcohol-fed rats 2. The compound reduced proteins involved in alcohol metabolism, including ADH1 and CYP2E1, and changed activity in the TCA cycle 2. But the compound is not mechano growth factor, and no MGF pathway was tested 2.
The second study looked at ginger-processed anemarrhenae rhizoma, an herb preparation 3. It identified chemical differences after processing and predicted that key components could bind to NF-kB and COX-2 proteins involved in inflammation 3. In a macrophage inflammation model, the preparation reduced TNF-alpha and IL-6 levels 3. This study also did not involve MGF 3.
MGF in exercise research
Mechano growth factor is a splice variant of the insulin-like growth factor 1 gene 4. Unlike the circulating form of IGF-1, MGF is produced locally when tissues are mechanically loaded, injured, or starved of oxygen 4. MGF carries a unique E-domain tail that lets it signal through pathways that are partly independent of the classic IGF-1 receptor 4. The new review includes MGF among the local IGF-1 isoforms it considers, but it does not separate MGF-specific results from the broader IGF-1 axis 1. Earlier research showed that androgen signaling can increase MGF expression in muscle as part of a broader anabolic program 7. So the review's placement is consistent with older data, but it does not add direct MGF evidence 1.
Direct MGF studies in repair and protection
Direct MGF research remains preclinical. In mice, a synthetic E-domain peptide from MGF reduced cell death and preserved heart function after an experimentally induced heart attack 8. In a rabbit model, MGF accelerated repair of the anterior cruciate ligament and improved movement of mechanically injured human ligament cells 9. In cartilage cells under low oxygen, pretreatment with an MGF E peptide improved cell proliferation and extracellular matrix production 10. In osteoarthritis models, MGF slowed joint degeneration in rabbits 11. In bone research, MGF promoted proliferation and osteogenic differentiation of rabbit mesenchymal stem cells 12 and enhanced bone-defect healing in rabbits 13. In the aging mouse brain, MGF increased the growth of new neurons in regions linked to memory and smell 14. MGF also promoted migration of human mesenchymal stem cells, which are used in tissue engineering 15. These studies provide the main body of direct MGF evidence, and none of them are human trials 4.
The naming and measuring gap
The limitation is that the new review names MGF without measuring it 1. The review's results describe the IGF-1 axis as a whole, and MGF is only one part of that axis 1. It cannot tell us whether low-load training changes MGF levels or activity in human muscle 1. The two other studies are not MGF research, so they cannot close that gap 2. For a reader, this means the new batch does not add MGF-specific findings 1.
What we still do not know
No controlled human efficacy trials for MGF exist 4. The direct MGF studies use cell cultures or animals, and safe duration, long-term risks, and interactions with common medications remain unstudied in humans 4. The new review does not provide MGF-specific measurements 1. The liver and herb studies do not involve MGF, so they cannot inform MGF's effects 2. For now, the direct MGF picture is unchanged: it is a local repair signal with animal and cell findings, but no human data 4.
This post describes what published studies did and found. It is research reporting, not medical advice, and it does not recommend use of any compound.
