AOD-9604 is a lab-made fragment of human growth hormone 1. In 2026, the animal studies still show fat loss without the insulin effects of full growth hormone, but human research stopped at early safety reports 2. The published work does not support strong human claims, and this post walks through what is actually there.

What AOD-9604 is

The compound is built from 16 amino acids: the tail end of human growth hormone, with an extra tyrosine added at the front 1. It was designed to keep growth hormone's fat-moving action while dropping the blood-sugar effects that full growth hormone carries 1. In lab tests, AOD-9604 does not bind to the growth hormone receptor and does not trigger receptor-driven cell growth 3. The development program reached phase two obesity testing by early 2002, but no approval ever followed 6. Today the compound appears mainly in anti-doping testing reports and in seizures of unregulated vials in the United States, Denmark, and Belgium 1.

Animal fat-loss findings

The strongest AOD-9604 data come from rodent studies done more than twenty years ago. Three studies stand out, each from the same small group of collaborating scientists 2.

StudyModel and treatmentDurationMain finding
Ng 2000 2Obese Zucker rats, 500 mcg/kg by mouth daily19 daysWeight gain was less than half that of controls; fat tissue showed more lipolytic activity
Heffernan 2001 3Obese mice, continuous treatment by pump14 daysLess weight gain, more fat oxidation, higher plasma glycerol, no rise in blood glucose
Heffernan 2001 9Obese and lean mice, continuous treatment14 daysReduced weight gain and restored beta-3 receptor RNA; knockout mice had a smaller chronic response

This information is for research only. Not intended for human use.

Across those studies, the same pattern appears: AOD-9604 increases fat breakdown and reduces weight gain in obese animals while leaving blood sugar and insulin alone 2. The studies are internally consistent, but they are not widely replicated by outside labs 2. The animal responses appeared within 14 to 19 days of continuous dosing 2. No human study has reproduced these effects.

Human research so far

Human evidence is confined to early reports. A 2003 abstract described favorable safety and tolerability in a phase two trial 10. A 2005 abstract said the orally active peptide "may aid weight reduction in obese subjects" 11. The published abstracts do not report dose, route, or regimen 6. Reviews through 2026 still list AOD-9604 among growth-hormone-axis peptides without strong human efficacy data 4. The evidence does not establish that AOD-9604 helps people lose weight. That means no one can say whether fat loss in people would be meaningful, because the human reports did not include numbers.

Why insulin effects stay quiet

Full growth hormone raises blood sugar and makes insulin less effective. AOD-9604 was designed to avoid that side effect. In obese mice, 14 days of treatment increased fat breakdown without raising blood glucose or suppressing insulin secretion 3. In obese rats, 19 days of oral treatment left insulin sensitivity normal on a euglycemic clamp (a test that measures how well insulin keeps blood sugar stable) 2. That fits the cell data: the peptide does not attach to the growth hormone receptor, so it does not trigger the same downstream actions 3.

Part of the fat effect appears to involve beta-3 adrenergic receptors, small switches on fat cells that help release stored fat 9. Chronic treatment in obese mice restored these receptors in fat tissue toward lean levels, and mice missing the receptor lost much of the long-term weight response 9. The working model is an early direct push on fat breakdown plus a later adjustment that keeps fat tissue responsive 2. The exact molecule the peptide acts on is still not identified 3.

The single joint study

One rabbit study looked beyond fat tissue. Researchers injected 0.25 milligrams of AOD-9604 into a knee osteoarthritis model each week, with or without 6 milligrams of hyaluronic acid 12. Over four to seven weeks, cartilage structure and walking ability improved compared with a saline injection, and the combination beat either treatment alone 12. That is the only joint evidence for AOD-9604. No human joint trial has been published 13.

Anti-doping and supply

AOD-9604 is prohibited in sport at all times under the WADA list as a growth hormone fragment 1. Testing laboratories can find it in urine down to 50 picograms per milliliter 1, and newer direct injection methods cover 50 to 500 picograms per milliliter 14. A stable metabolite extends the detection window beyond the parent peptide 1. Dried blood spot workflows can also detect the peptide and stay stable long-term without refrigeration 15. The peptide does not interfere with the WADA test for full growth hormone, so it neither masks nor confuses that result 16.

Outside sport, unregulated supply is a documented problem. AOD-9604 has been identified in confiscated vials in the United States 1, in peptide seizures in Denmark between 2007 and 2013 7, and in seized pharmaceutical preparations in Belgium 8. Those analyses show the peptide is present in gray-market products, but they do not verify that the labeled dose matches what is inside 8.

What we still do not know

The biggest gap is human efficacy. No published peer-reviewed trial has tested whether AOD-9604 leads to meaningful weight loss in people 4. Reviews in 2026 note that gray-market use of growth-hormone-axis peptides runs ahead of clinical evidence 4. Long-term and cumulative safety is also unknown, because human exposure in the development program was short 6. The exact molecular target for fat breakdown remains unidentified 3. The joint evidence rests on a single rabbit study, so it cannot be generalized 12. Supply quality is another unresolved problem: confiscated vials in Denmark and Belgium have been identified as AOD-9604, but their composition and dose accuracy are not verified 7. Until better evidence exists, the animal findings are the main thing the literature can support.

This post describes what published studies did and found. It is research reporting, not medical advice, and it does not recommend use of any compound.
