DSIP studies in 2026 still show mixed sleep evidence
The small human trials are decades old and split: a 7-night intravenous course improved severe insomnia, while a 3-night trial saw only weak changes. Animal work shows sleep, pain, and seizure effects, but no DSIP receptor has ever been identified.

Does a peptide first found in sleeping rabbits change sleep in people? The human evidence, mostly from small intravenous trials conducted decades ago, is split. One seven-night course improved severe insomnia, while a three-night trial saw only weak changes. This post reviews the DSIP research summary and what the studies show as of October 2026.
What the human sleep trials found
The strongest evidence comes from one placebo-controlled trial and a separate 3-night trial in people with chronic insomnia. In a seven-night study of 14 people, nightly intravenous DSIP improved night sleep from the first dose, and the benefit persisted through the first night after treatment ended 1. Daytime alertness and performance also returned to normal 1. A separate three-night trial in 16 people used 25 nanomoles per kilogram, roughly 21 micrograms per kilogram, and found only weak improvements in sleep efficiency and how long it took to fall asleep 2. The authors concluded that short-term DSIP was unlikely to be of major therapeutic benefit 2. A 1981 report also described improved sleep in people with insomnia 3.
| Study | People | Dose and route | What changed |
|---|---|---|---|
| 7-night insomnia trial 1 | 14 adults with severe chronic insomnia | Nightly intravenous, 7 nights | Sleep improved from first dose; daytime function normalized |
| 3-night insomnia trial 2 | 16 adults with chronic insomnia | 25 nmol/kg intravenous, 3 nights | Weak improvements in sleep efficiency and latency |
| Chronic pain pilot 4 | 7 adults with chronic pain | Intravenous daily for 5 days, then spaced injections | Pain reduced in 6 of 7; mood improved |

The pain pilot did not measure sleep as a main endpoint, but it is part of the small human evidence base.
Why the insomnia results split
The studies used different schedules and possibly different timing. DSIP leaves the blood fast, with a half-life of about two to four minutes in dogs, rodents, and monkeys 5. Evening dosing aligns with the body's natural wind-down, and afternoon dosing has been suggested as one reason the three-night trial underperformed 2. Animal work also shows a bell-shaped dose-response curve, meaning doses above the effective window can become less active 6. So the split may be about when and how much, not whether the peptide can affect sleep at all.
What animal models add
A 2026 review groups DSIP with Epithalon and pinealon as recovery-enhancing peptides that act on circadian and mitochondrial regulators 7. Animal research is more consistent on sleep. Rabbits show more light non-REM sleep and cats show more REM sleep, both with bell-shaped dose-response curves 6. A fusion version of DSIP that crosses into the brain restored neurotransmitter balance and sleep in mice with insomnia 8. Under simulated high altitude, a phosphorylated form improved non-REM (NREM) and REM sleep and spatial memory in rats 9.
Beyond sleep, animal models report several effects. DSIP reduced pain in mice and increased release of met-enkephalin, a natural opioid-like signal 10. In a small human pain pilot, six of seven people reported less pain after intravenous treatment 4. DSIP also reduced seizure intensity in a drug-resistant seizure model 12. After experimental stroke, intranasal DSIP sped motor recovery in rats, although the reduction in stroke size was not statistically significant 13. A separate ischemia study found a hard constraint: giving DSIP or its analogue during active vessel blockage was fatal in animals, while giving it after blood flow returned reduced tissue damage 14. In aging mice, a DSIP-containing preparation called Deltaran improved biomarkers, extended lifespan, and reduced spontaneous tumor incidence 15.
The missing receptor
One reason the human picture is hard to read is that nobody has found a DSIP receptor, gene, or precursor 17. DSIP is best described as a central neuromodulator rather than a key fitting one lock. The intact peptide crosses the blood-brain barrier by passive diffusion 18, so fast clearance does not rule out brain effects, but it does make timing important.
What the mechanism studies show is indirect. DSIP inhibits the release of somatostatin, a brake on several pathways, through a dopamine-related route 19. It also increases met-enkephalin release, and naloxone, an opioid blocker, reverses several DSIP effects in rats 10. In stress models, DSIP blunts CRF (a stress signal)-triggered release of corticosterone (a rodent stress hormone) in rats, pointing to an action in the pituitary gland 21, but a healthy human study found no change in the stress hormones ACTH or cortisol after DSIP 22.
Safety and legal status
The small human trials and the pain pilot reported no serious adverse events 1. Sedation is the most common effect and is also the intended direction 1. Next-day performance was not impaired in the seven-night trial; daytime function normalized 1. Formal adverse-event collection was not performed, so side-effect frequencies cannot be quantified 1.
The clearest safety signal comes from ischemia studies. DSIP or its analogue KND during active coronary or cerebral artery occlusion caused 100% mortality in animals, while reperfusion-phase dosing reduced infarct size 14. Research guidance therefore treats active myocardial ischemia, unstable angina, acute stroke, and a transient ischemic attack (TIA, a brief stroke) as hard stops 14. A 2021 survey found DSIP in illegally distributed peptide products 23.
What we still do not know
The biggest gap is the missing signaling system. No receptor or gene has been identified 17, so it is hard to say how the peptide works or what else it might do. Human exposure beyond roughly three weeks is unstudied 1. The optimal route, dose, and duration are open. Tolerance was not observed in the short trials, but community reports describe fading benefit, and no long-term human data exist 1. Community stacks often add BPC-157, ipamorelin, or CJC-1295, but there are no human combination trials. The grey-market supply is also unverified 23. In the research so far, DSIP has mixed human sleep evidence, plausible animal effects, and a missing receptor that leaves many questions open.
This post describes what published studies did and found. It is research reporting, not medical advice, and it does not recommend use of any compound.
